钒氧羧酸类配合物的合成及降糖活性研究

摘    要:目的设计合成了双(α-噻吩甲酸)氧钒二甘醇酸氧钒和双(1,1-环丁烷二羧酸)氧钒3个新型的有机羧酸氧钒配合物,并进行降糖活性研究.方法应用元素分析、红外光谱、紫外光谱、质谱和核磁共振氢谱对所合成配合物的结构进行初步确证.在实验性糖尿病动物模型上研究3个配合物的降血糖作用和初步的毒性反应.结果成功合成了3个羧酸氧钒配合物,其中二甘醇酸氧钒(VO—DA)灌胃给药明显降低四氧嘧啶性糖尿病小鼠的餐后血糖和空腹血糖。3个化合物均有一定的胃肠道毒副反应.结论合成的3个配合物元素分析数据、结构参数与计算值相符合,表明所合成的3个配合物为目标配合物.配合物VO—DA初步显示出降糖活性.

Abstract:Objective To design and synthesize oxovanadium carboxylate coordination compounds,and to their hypoglycemic effects in alloxan-diabetic mice.Methods The chemical structure of the complexes was determined by elemental analysis,IR,UV,MS and H-NMR spectra.The effects of the compounds on the blood glucose concentrations and preliminary toxicity were investigated in alloxan-induced diabetic mice.Results Three oxovanadium carboxylate coordination compounds have been synthesized successfully.After 3 weeks administration,diglycollato oxovanadium(VO-DA) significantly reduced the fasting and post-prandial blood glucose level in alloxan-diabetic mice.All the compounds have some adverse effects of gastrointestinal tract.Conclusion VO-DA certainly has anti-diabetic effect in alloxan-induced diabetic mice.
