补肾方剂对糖尿病大鼠纯钛种植体骨结合影响的组织学研究

摘    要:目的通过建立糖尿病大鼠骨内种植模型,研究补肾方剂对糖尿病大鼠纯钛种植体骨结合的影响。方法18只Wistar大鼠,4月龄,雄性,随机分为三组:糖尿病组、糖尿病补肾方剂治疗组、对照组,每组6只。糖尿病组、糖尿病补肾方剂治疗组空腹一次性腹腔注射STZ溶液(45mg/kg),注射STZ三天后,大鼠空腹血糖高于16.65mmol/L,尿糖(+++),糖尿病大鼠模型建立成功。对照组大鼠空腹腹腔注射等量生理盐水。三组动物,于左侧胫骨近中干骺端外侧制备直径1.0mm的种植孔,植入种植体,初期稳固;种植体植入术后第一天起,治疗组用补肾方剂灌胃(用量:补肾方剂90g水煎浓缩成1g/ml生药浓度药液每只动物1.5ml 2次/日),对照组和糖尿病组自由饮食。种植术后12周,取下带种植体的胫骨,制作HE染色组织切片,采用光镜和骨组织形态计量学观察。结果光镜观察发现,补肾方剂治疗组结合骨板连续性较好、较厚,表面光滑,周边可见大量的成骨细胞。骨组织形态计量学参数比较发现,治疗组结合骨板的宽度、种植区骨细胞个数及非种植区骨细胞个数与对照组无显著差异(P〉0.05);治疗组种植区成骨细胞个数显著高于对照组(P〈0.05);糖尿病组结合骨板的宽度、种植区成骨细胞个数、种植区骨细胞个数及非种植区骨细胞个数均显著低于治疗组(P〈0.05)。结论补肾方剂可通过控制糖尿病和调节骨代谢有效促进和提高纯钛种植体的骨结合。

Histological study on titanium implants osseointegration in diabetic rats treated by tonifying kidney recipes

Abstract:Objective To study the effect of tonifying kidney recipes (TKR) on osseointegration of titanium implants in diabetic rats by establishing endosseous implant models of the diabetic rats. Methods Eighteen Wistar rats, 4 months old, male, were used in this study and divided into 3 groups of 6 rats each. The rats with empty stomach were injected by STZ solution (45mg/kg) intraperitoneally of diabetes group and TKR group. Three days after injection, the blood - glucose level was higher than 16.65mmol/L, and urine sugar was positive (+++), which suggested the success of diabetes models established, simultaneously the rats were injected by normal saline intraperitoneally of the control group. In the three groups, the implant beds in 1.0mm diameter were made in the off- side of the mesio - occlusion metaphysis of the left tibia, and the implantation was performed. The first day postoperatively, TKR was administered intragastrieally in the TKR group, while the control group and diabetes group did not administer any medicine in diet. Twelve weeks postoperatively, samples of rats were obtained, and the sections (HE stainning) were made and observed by light microscope and bone histomorphometric method. Results The results of light microscope observation: In TKR group, the combined bone lamella was relatively thick with relatively good continuity and a smooth surface, around which lots of osteoblasts could be seen, with the density of osteocytes relatively high. Bone histomorphometric results found that the combined bone lamella width, the numbers of the implant area osteocytes and the non - implant area osteocytes in TKR group were no significantly different from those of the control group ( P 〉 0.05 ), whereas the numbers of the implant area osteoblasts in TKR group were significant higher than those in the control group.
