即刻早期基因c-fos,c-jun在缺血再灌注大鼠肝脏中的表达

目的研究即刻早期基因家族中c-fos、c-jun基因在缺血再灌注肝脏中的表达情况,探讨其与缺血再灌注损伤发生的关系。方法 SD大鼠随机分为假手术组、缺血再灌注后30min组和缺血再灌注后120min组。全自动生化分析仪测定血清ALT、AST水平,光镜观察肝组织形态学改变,TUNEL法测定肝细胞凋亡指数。实时荧光定量PCR（realtimePCR）法测定组织中c-fos及c-jun的表达情况。结果肝脏缺血再灌注损伤后血清ALT、AST逐步上升（P〈0.05）。病理改变显著,细胞凋亡现象逐步显现。c-fos及c-jun的表达水平,在再灌注后30min明显升高（P〈0.05）,再灌注2h后表达水平有所回落,但仍显著高于假手术组（P〈0.05）。结论 c-fos及c-jun基因参与肝脏缺血再灌注损伤的发生,并可能与肝细胞凋亡与再生的过程相关。

The expression of immediate early genes(IEGs) in liver of ischemia reperfusion rats

Aim To investigate the expression of c-fos and c-jun gene of IEGs family in liver ischemia reperfusion（I/R） injury.Methods SD rats were randomly divided into shame operation group,30min after reperfusion group and 120min after reperfusion group.The changes in hepatic histomorphology,enzymology and the apoptosis index were analysised.The experssion level of c-fos and c-jun were measured by real time PCR.Results After ischemia reperfusion,the level of enzymology and apoptotic index was increased gradually（P0.05）.The expression of c-fos and c-jun was notable 30min（P0.05） after reperfusion and decreased 120 min after reperfusion,but was higher than that of OS group（P 0.05）.Conclusion The c-fos and c-jun genes involve in the development of liver I/R.
