金丝桃苷对离体缺氧/缺糖再灌注脑损伤的保护作用

摘    要: 目的研究金丝桃苷对离体缺氧缺糖/再灌注脑损伤的保护作用。方法小鼠离体脑片缺氧缺糖/再灌注(OGD/RP)诱导的模型上,以2,3,5-三苯基氯化四氮唑(TTC)产物formazan定量测定脑片活性,同时计数单位面积脑片皮层和纹状体区的存活神经元。新生大鼠原代培养神经元OGD/RP损伤后测定乳酸脱氢酶(LDH)释放和噻唑蓝(MTT)液比色吸光度,评价神经元活性。观察不同浓度的金丝桃苷对于离体脑损伤的保护作用。结果金丝桃苷可有效减轻OGD/RP对脑片的缺血性损伤,并对于OGD/RP原代神经元具有显著的保护作用。结论金丝桃苷对离体缺氧缺糖/再灌注脑损伤具有明显的保护作用,此作用可能与其抗自由基生成、抑制Ca2+内流及脂质过氧化物形成有关。

Abstract:OBJECTIVE To investigate the protective effects of hyperin on brain injury induced by oxygen-glucose deprivation/reperfusion (OGD/RP) in vitro. METHODS After oxygen-glucose deprivation for 15 min followed by reperfusion for 30 min, brain slices were stained with 2,3,5-trihenylterzolium chloride (TTC). TTC red product formazan was measured to evaluate the viability of brain slices.Survival neural cells of the brain slice were also counted. Lactate dehydrogenase (LDH) release and MTT reduction in neuron cultures were measured after OGD/RP injury. Hyperin at the different concentration was added directly into the aCSF and neuron-culture solutions during the course of OGD/RP. RESULTS The formazan of brain slices were decreased after OGD/RP, and hyperin(1-10 μmol·L-1) markedly attenuated OGD/RP induced injury in cortex and striatum. Hyperin( 10-100μmol· L-1) also effectively increased the tolerance of neonatal rat's neurons to OGD/RP injury. CONCLUSION Hyperin has protective effects on cerebral injury induced by OGD/RP, which may due to the inhibition of free radical production, Ca2+ influx and lipid peroxidation production.
