圆弧青霉提取物诱发人淋巴细胞SCEs的研究及TLC分析

摘    要:采用从食管癌高发区河南省林县分离出的4株程序外DNA合成(UnscheduledDNA Syntesis,UDS)与DNA合成抑制(DNA Synthesis Inhibition,DSI)试验阳性、Ames和枯草杆菌DNA 重组试验阴性的圆弧青霉提取物,诱发正常人外周血淋巴细胞姊妹染色单体交换(Sister Chr matid Exchanges,SCEs)。结果表明,4株圆弧青霉提取物均能诱发SCEs频率升高(P<0.001),并呈明显的剂量效应关系。同时,意外地发现,对PHA刺激后的T淋巴细胞增殖周期,有明显的剂量效应阻滞作用(P<0.01)。提示诱变剂有可能同时也攻击免疫细胞,起到免疫抑制剂的作用。TLC分析初步证实,4株圆弧青霉提取物成分近似,解释了体外生物学短期诱变试验为何得到基本一致的结果。

Abstract:The extracts of 4 strains of P. cyclopium, which had induced positive UDSand DSI in human amnion FL cell line and negative Ames test as well as Bacillussubtilis rec-assay before, could induce high frequency of SCEs in the lymphocytesof normal human peripheral blood (P<0.001), and there was obvious relationwith dosage effect. At the same time. evident dosage effect blocking of cell divi-sion cycles of T lympho ytes irritated by PHA was unexpectively found (P<0.01).The extracts of the 4 strains of P. cyclopium had nearly the same compositionpreliminaryly confirmed by TLC. This explained the reason why the results in biol-ogical mutagenic assays were similar. Therefore, in the extracts there definitelywas mutagen(s) that could damage DNA in human cells. But the carcinogenisisof the extracts needs further to be studied. In tumor etiology, it is worthwhile tonotice that mutagens may attack the DNA of active immunologic cells, ie theyplay the part of immunosuppresive agents.
