乳癌组织中TP、TS mRNA及蛋白的表达

摘    要:目的:探讨乳癌组织中TP、TS mRNA及蛋白表达水平及其与临床病理特征、预后的关系。方法:采用定量PCR及免疫组化技术检测经“微选”的9例正常乳腺组织和86例乳癌组织TP、TS的mRNA和蛋白表达水平。结果:TP、TS mRNA在乳癌及正常组织中的表达水平存在明显的个体差异。TP、TS的mRNA与蛋白表达无相关性(r=0.147,P=0.114和r=0.111,P=0.114)。TP、TS mRNA或蛋白表达与肿瘤体积、淋巴结转移、组织学分级、临床分期均无关。10a随访结果显示TP表达与预后无关,而高TS mRNA或蛋白表达患者总生存期短于低表达者,显示较差的临床预后。结论:乳癌和正常组织巾TP、TS的mRNA、蛋白表达水平变化较大,TS检测可作为判断乳癌预后的指标之一。

Study on relationships between mRNA, protein expression of thymidine phosphorylase, thymidylate synthase and clinicopathology, the prognostic significance in human breast carcinomas
Abstract:Aim: To study the relationships between mRNA , protein expression levels of thymidine phosphorylase (TP) , thymidylate synthase (TS) and the clinic pathological features, prognosis in human breast carcinomars. Methods: mRNA, protein expression levels of TP, TS were detected by real-time quantitative PCR and immunohistochemistry in a series of 86 micro-selected breast carcinomas tissues and 9 micro-selected normal breast tissues. Results: The expressions of TP, TS mRNA had evident individual differences among breast carcinomars and normal breast tissues. There was no association between mRNA and protein expression levels of TP, TS. The levels of TP,TS mRNA or protein expression had no association with the tumor size, the metastasis of lymph node, pathological grade and clinical stage. Ten year follow-up analysis showed no association between TP expression and prognosis. High level of TS mRNA or protein expression, however, was associated with a worse clinical prognosis. Conclusion : The levels of TP, TS mRNA and protein expressions were highly variable in breast carcinomars and normal breast tissues, and TS expression may be a prognostic marker in breast carcinoma patients.
