| 摘 要： | 目的：探讨体外诱导骨髓基质细胞向心肌细胞转化的条件。方法：分离大鼠骨髓，体外培养、传代得到骨髓基质细胞，观察不同浓度的诱导剂5-氮胞苷对骨髓基质细胞的生长和诱导作用，进行形态学和免疫组织化学鉴定。结果：5-氮胞苷诱导骨髓基质细胞转化为心肌样细胞，在诱导后2周转化率为29％。心肌特异性肌钙蛋白T(TnT)免疫组织化学染色阳性。结论：MSCs在体外条件下经5-氮胞苷诱导可分化成心肌样细胞。

| Abstract: | Objective To study the differentiation of mesenchymal stem cells (MSCs) into cardiomyocytes by 5-azacytidine (5-aza) in vitro. Methods Rat MSCs were isolated and cultured,then co-cultured with 5-aza at different concentration for 24 h. After cultured for additional 2 weeks,MSCs were identified by means of immunochemistry technique. Results A part of the cells affected by 5-aza had morphological changes and 29% of it was stained positively for Tn T. Conclusion MSCs can be induced into cardiomyocytes by 5-aza in vitro. |
