p53基因突变与人食管癌转移的关系研究

目的：探讨ｐ５３基因突变与人食管癌淋巴结转移的关系。方法：采用聚合酶链反应－单链构象多态性（ＰＣＲ－ＳＳＣＰ）结合银染技术，对４７例食管瘤组织中ｐ５３ 基因第５～８外显子进行了突变分析。结果：４７例食管癌标本中，ｐ５３基因第５～８外显子出现突变者２０例，其突变率为４２．６％；其中２２例伴有淋巴结转移的原发癌中１３例出现了突变（５９．１％），其与无淋巴结转移组（２８．０％，７／２５）相比有显著性差异（Ｐ＜０．０５）。结论：ｐ５３基因突变与人食管癌的转移可能关系密切。

A study on the relationship between p53 gene mutation and lymph node metastasis of hmnan esophageal carcinoma

Objective: To discuss the relationship between p53 gene mutation and lymph node metastasis of human esophageal carcinoma.Methods: PCR-SSCP methods and silver staining technique were used to analyse the mutations of p53 gene exons 5-8 in 47 cases of esophageal carcinoma samples. Results: Mutation in exons 5-8 of the p53 gene were found in 20 cases out of 47 patients with esophageal carcinoma. The total mutation rate was 42. 6% (20/47) .Mutation of p53 gene were detected 13 of 22 cases with lymph node metastasis(59. 1 %, 13/22). The incidence of mutation of p53 gene in the cases of lymph node metastasis was obviously higher than that in patients without metastasis (28. 0%,7/25) (P < 0.05). Conclusion:These results suggest that p53 gene mutation were related to metastasis of human esophageal carcinoma.
