环氧化酶2抑制剂对卡介苗激活的树突状细胞介导的体外抗膀胱肿瘤免疫效应的影响

摘    要:目的体外实验研究环氧化酶2(Cox-2)抑制剂对卡介苗(BCG)感染的人脐血源性树突状细胞(CBDC)介导的抗膀胱肿瘤免疫效应的影响。方法从人脐带血中分离和体外诱导培养得到树突状细胞(DC)。采用BCG感染DC,并与前列腺素E2(PGE2)、塞来昔布(Celecoxib)等共培养,检测各组DC的混合淋巴细胞反应性(MLR)及DC激活的细胞毒T淋巴细胞(CTL)对人膀胱肿瘤细胞株T24的细胞毒活性。结果 MTT实验显示Celecoxib显著增强、而PGE2则明显抑制经BCG感染的DC的MLR;乳酸脱氢酶(LDH)释放实验表明,Celecoxib显著增强、而PGE2则明显抑制经BCG感染的DC激活CTL的抗膀胱肿瘤细胞毒活性。结论选择性Cox-2抑制剂Cele-coxib能够增强BCG激活的DC介导的体外抗肿瘤免疫效应。
