葛仙汤治疗大鼠溃疡性结肠炎

目的:探讨葛仙汤对大鼠溃疡性结肠炎的治疗作用及其机制.方法:将SD大鼠按均衡随机原则分为正常对照组、模型对照组和葛仙汤低、中、高剂量组,采用大肠杆菌加乙酸免疫复合法造成大鼠溃疡性结肠炎模型.连续给药21 d,观察动物的一般情况,进行疾病活动指数(DAI)评估、结肠病理学评分(HS),检测血清中IL-6、IL-10含量.结果:葛仙汤低、中、高剂量3个治疗组DAI和HS均明显低于模型对照组(P<0.05或P<0.01),IL-6含量与模型对照组比较均明显减少(P<0.01);中剂量组IL-6含量明显低于高、低剂量组(P<0.05).3个治疗组IL-10均较模型对照组明显升高(P<0.01);中、低剂量组IL-10明显高于高剂量组(P<0.05).结论:低、中、高剂量葛仙汤均能缓解溃疡性结肠炎大鼠的症状,减轻其结肠病理损伤,促进溃疡面愈合,其中以中剂量效果为更好.

Gexiantang on Treatment of Ulcerative Colitis in Rats

Objective To investigate the effect of Gexiantang(GXT) on treatment of ulcerative colitis(UC) in rats and the mechanism.Method Sprague Dawleg(SD) rats were in equilibrium divided randomly into 5 groups,including control group,model group,low dosage GXT group,medium dosage group and high dosage group.UC rat model was established by utilizing combined Escherichia coli and acetate acid.After 21 days treat ment,observation on rats,regular situation was done with evaluating UC rats by disease activity index(DAI),his topathological score(HS) and double antibody sandwich ABC-ELISA method in the detection of serum contents of IL-6,IL-10.Results Improvement in low dosage GXT group,medium dosage GXT group and high dosage GXT group was shown comparing to the model group.Three treatment groups showed significantly lower expres sion than that of the model group in DAI and HS(P < 0.05 or P < 0.01).IL-6 levels in 3 treatment groups were significantly lower than that in the model group(P<0.01).Medium dosage GXT group IL-6 level showed significantly lower than that in high and low dosage GXT groups(P < 0.05).In the 3 treatment groups IL-10 level showed higher than that in the model group(P < 0.01).IL-10 level in medium and low dosage GXT groups showed significantly higher expression than that in high dosage GXT group.Conclusion It is suggest ed that low dosage,medium dosage and high dosage GXT are all able to relieve symptoms of UC rats,to reduce colonic pathological damage,and to promote the ulcer surface healing.These are possibly related to IL6 inhibi tion of inflammatory factors releasing and promoting anti-inflammatory cytokines IL-10 generation from GXT.Medium dosage GXT on effect of IL-6 and IL-10 is stronger than its higher dosage.
