同型半胱氨酸致血管内皮细胞DNA损伤的作用

摘    要:[目的]探讨同型半胱氨酸(HCY)对血管内皮细胞的DNA损伤作用。[方法]体外培养鼠主动脉内皮细胞,随机分为正常对照组和HCY9种浓度组,MTT法检测细胞毒作用,彗星试验检测DNA损伤情况,取培养液测定丙二醛(MDA)含量和超氧化物歧化酶(SOD)活力。[结果](1)对照组细胞呈单层鹅卵石样,紧密排列。1、2.5、5mmol/L HCY组细胞无明显变化,而自10mmol/L起,HCY组细胞排列杂乱,聚集成团。MTT法显示细胞生长明显受抑制,抑制率达20%;(2)彗星试验显示自5mmol/L HCY起尾长、尾矩和Olive尾矩明显增加;(3)自10mmol/L起HCY培养液中MDA含量高于对照组,SOD活力低于对照组。[结论]HCY具有明显的细胞毒作用,其机制可能与HCY直接造成DNA损伤和促进氧化应激有关
