亚低温联合盐酸法舒地尔对创伤性脑损伤后Bcl-2和Bax表达的影响

摘    要:目的:探讨亚低温联合脑血管扩张药盐酸法舒地尔对脑外伤(TBI)大鼠脑细胞Bcl-2、Bax蛋白表达的影响及其对外伤后受损害脑组织的保护作用。方法:30只SD大鼠随机分为假手术组(Sham)、创伤性脑损伤组(TBI)、亚低温治疗组(MHT)、法舒地尔治疗组(FSD)及亚低温联合法舒地尔组(MHT+FSD),每组6只。Allen's改良法制作海马细胞凋亡的模型,予以亚低温及亚低温联合法舒地尔治疗。HE染色观察海马细胞形态学的变化;免疫组化法检测神经细胞Bcl-2和Bax蛋白表达的变化。结果:(1)HE染色。MHT组与FSD组海马细胞均较TBI组形态规则,胞核浓染、固缩的现象明显减少。与MHT组及FSD组相比,MHT+FSD组细胞层次更多,排列也更为整齐。(2)免疫组化检测。与TBI组相比,MHT组及FSD组Bcl-2蛋白达表水平显著增高(P〈0.05),Bax的表达明显降低(P〈0.05);与MHT组及FSD组相比,MHT+FSD组Bcl-2蛋白表达显著增强(P〈0.05),Bax的表达显著降低(P〈0.05)。结论:亚低温联合法舒地尔治疗可通过有效抑制TBI后细胞凋亡而发挥脑保护作用。

Abstract:Objective:To investigate effects of mild hypothermia (MHT) combined with fasudil (MHT + FSD) on expression of Bcl -2 and Bax in rats after traumatic brain injury (TBI), and explore this therapeutic regimen protection for brain tissue against TBI. Methods:Thirty SD rats were randomly divided into five groups: sham group, TBI group, MHT group, FSD group and MHT + FSD group (n = 6). The models of hippocampus cell apoptosis in rats were established by improved Allen's method. HE staining was used to observe the changes of cell morphology, immuno - histochemical staining to detect expression of Bcl-2 and Bax. Resuits:HE staining showed that the hippocampus cells in MHT and FSD groups presented more regular appearance with dense nucleus and fewer karyopycnosis than those in MHT + FSD group, moreover, more cellular layers and well arranged cells were observed in MHT + FSD group. The immunohistochemistry staining demonstrated that the expression of Bcl-2 was significantly stronger and Bax expression was markedly lower in hippocampus CA3 region in MHT + FSD group than that in MHT and FSD groups. Conclusions : Combination therapy of mild hypothermia and fasudil may strengthen the brain protection through inhibition of cell apoptosis following traumatic brain injury.
