青钱柳提取物清除羟自由基和抑制脂质过氧化的作用

摘    要:目的研究五种青钱柳提取物(CPS)清除羟自由基和抑制脂质过氧化作用。方法采用组织匀浆MDA比色法测定不同浓度的CPS对小鼠肝脏自发性脂质过氧化的作用。利用Fenton反应产生.OH,研究CPS对羟自由基的清除作用,计算半清除率(IC50)。结果五种提取物均明显的具有抗脂质过氧化作用,在浓度为1.0mg.ml-1时,对小鼠肝组织自发性MDA的抑制率大小为:正丁醇>石油醚>多糖>水>乙酸乙酯。CPS对.OH自由基IC50大小为:水>石油醚>正丁醇>多糖>乙酸乙酯。结论五种青钱柳提取物均有较强的体外抗氧化活性,且活性与浓度呈明显的量效关系。

Abstract:Objective To study the effect of five kinds Cyclocarya paliurus(Bata1. ) Iljinskaya Extracts(CPS) on scavenging hydroxy radical and stopping lipid peroxidation. Methods The effect of different density CPS on mice liver spontaneous lipid peroxidation were detected by tissue homogenate MDA chromatometry. Invest the hydroxy radical scavenge effect of CPS which produced by Fenton reaction. Then calculated the 50% inhibiting concentration (IC50). Results The experiments showed that the five kinds extracts had obvious effects on anti-lipid peroxidation. At the concentration of 1.0mg. ml^-1 ,it was found that the effect from strong to weak on MDA generation spontaneously in liver of mice was N-butanol,Petroleum ether, polysaccharide, water,Ethyl acetate, the potency from strong to weak of scavenging free radicals IC50 was water, Petroleum ether, N-butanol, polysaccharide, Ethyl acetate. Conclusion Five kinds Cyelocarya paliurus(Bata1. ) Iljinskaya Extracts have strong vitro function on antioxidant activity. There was obviously dose-effect relationship between activity and concentration.
