大鼠断肢再植后骨骼肌缺血再灌注损伤的保护

摘    要:目的 :研究断肢再植后离断肢体骨骼肌缺血再灌注损伤的保护。方法 :建立大鼠后肢的断肢再植模型 ,再植前应用灌注液对实验组离断肢体进行灌注 ,然后进行再植。缺血再灌注组不进行灌注直接通血 ,于通血后 6h取材 ,测定骨骼肌丙二醛 ( MDA)、超氧化物歧化酶 ( SOD)、线粒体ATP酶活性以及胫前肌含水量 ,观察骨骼肌损伤情况。结果 :灌注液处理组胫前肌含水量为( 77.86± 0 .75 ) %,ATP为 ( 0 .2 1± 0 .0 5 ) mmol· g-1· h-1 ,MDA为 ( 5 .62± 0 .35 ) μmol· g-1 ,SOD为 ( 2 7.41± 3.77) NU·mg-1 ,均较缺血再灌注组有明显改善。结论 :断肢再植前应用灌注液对离断肢体进行灌注 ,可以减轻再植肢体中骨骼肌的缺血再灌注损伤

Study on Prevention and Cure of Skeletal Muscle′s Ischemia Reperfusion Injury After Limb Replantation in Rats
Abstract:Objective:To study the protection of skeletal muscle′s ischemia reperfusion injury (IR) after limb replantation. Methods:After establishing the amputated replantation models of hindlimbs in rats, the experimental group was perfused with perfusion solution before replantation, but the IR group was not perfused. 6 hours later, the contents of malondialdehyde (MDA), superoxide dismutase (SOD), ATP enzyme of mitochondrial of skeletal muscle and water content of the tibialis anterior were measured. Results:In the experimental group, the water content of the tibialis anterior was (77.86±0.75)%, ATP enzyme of mitochondrial of skeletal muscle was (0.21±0.05) mmol·g -1 ·h -1 ,the contents of malondialdehyde (MDA) was (5.62±0.35)μmol·g -1 , the contents of superoxide dismutase (SOD) was (27.41±3.77) NU·mg -1 . The improvement was significant comparing with the IR group. Conclusion:The IR of skeletal muscle may be relieved if the amputated limbs are perfused with perfusion solution before replantation.
