| 摘 要： | 目的：建立并优化人骨髓间充质干细胞(mesenchymal stem cells，MSCs)分离、消化、传代扩增等培养方法．方法：以Percoil(1．073g／mL)、Ficoll(1．077g／mE)和A液(笔者配制)3种分离介质分离MSCs．传代时以不同浓度胰蛋白酶对MSCs进行消化．结果：A液对MSCs的分离效果最佳，不同蛋白酶液对MSCs消化能力不同。结论：建立了一套稳定、低耗、有效的MSCs培养方法。

| Abstract: | Objective:To establish and optimize a method for isolation,digest,subculture and expansion of mesenchymal stem cells (MSCs) from human bone marrow in vitro.Methods:MSCs were isolated with 3 isolation medium,1.073g/mL percoll,1.077g/mL Ficoll and Solution A (author offer).MSCs were digested and subcultured with different concentration trypsase or EDTA.Results:Solution A is the best to isolate MSCs.Different concentration trypsase make different effect for digest of MSCs.Conclusion: A stable,less expensive and efficient method for cultivation of MSCs from human has been established. |
