miR-539对乳腺癌细胞增殖与凋亡的调控作用研究

摘    要:目的:研究miR-539对乳腺癌细胞增殖和凋亡的调控作用。方法采用荧光定量PCR检测乳腺癌细胞(MCF-7、MDA-MB-453、ZR-75-30)和正常乳腺上皮细胞(MCF-10A)中miR-539表达的差异;通过转染miR-539-mim-ics在MCF-7细胞中上调miR-539的表达,并通过MTT和TUNEL实验检测miR-539对乳腺癌细胞增殖和凋亡的影响。结果与正常乳腺上皮细胞相比,乳腺癌细胞中miR-539的表达显著降低( P<0.01)。与空白对照组和阴性对照组相比,过表达miR-539能显著降低过表达组乳腺癌MCF-7细胞的增殖能力并增加其对地塞米松诱导凋亡的敏感性(P<0.01)。结论 miR-539能够抑制乳腺癌细胞增殖并促进其凋亡,有望作为乳腺癌基因治疗的靶点。

Regulated Role of miR-539 on Proliferation and Apoptosis in Breast Cancer Cells
Abstract:Objective To investigate regulation function of microRNA-539 (miR-539) on proliferation and ap-optosis of breast cancer cells. Methods Expressions of miR-539 in breast cancer cells (MCF-7, MDA-MB-453 and ZR-75-30) and normal breast epithelial cells (MCF-10A) were detected by fluorescent quantitation polymerase chain reaction ( PCR) . miR-539 expression was up-regulated by transfecting miR-539-mimics into MCF-7 cells, and then effects of miR-539 on proliferation and apoptosis of breast cancer cells were detected by MTT assay and TUNEL experiment. Re-sults miR-539 expression in breast cancer cells was significantly decreased compared with that in normal breast epitheli-al cells (P<0. 01). Proliferation ability of MCF-7 breast cancer cells was significantly decreased, and its sensitivity to apoptosis induced by Dexamethasone was increased in over-expressed miR-539 group compared with those in control and negative groups (P<0. 01). Conclusion MiR-539 can inhibit proliferation and promote its apoptosis of breast cancer cells, which may be used as a target for gene therapy of breast cancer.
