医用创面胶和吻合胶对小鼠成纤维细胞L929毒性的研究

摘    要:目的 探讨医用创面胶和吻合胶对小鼠成纤维细胞的间接毒性.方法 通过小鼠成纤维细胞L929检测两种胶的细胞毒性.采用CCK-8法检测两种医用胶对细胞活性的影响、细胞DAPI荧光染色检测对细胞分裂生长的影响、FITC/PI凋亡染色检测对细胞凋亡的影响.结果 创面胶组和吻合胶组小鼠成纤维细胞L929活性比较差异无统计学意义(P>0.05),但均低于对照组(P<0.05).3组荧光染色细胞计数、总面积占比和正常细胞存活率比较差异无统计学意义(P>0.05).结论 医用创面胶和吻合胶能够较明显降低周边毗邻细胞活性且降低程度相当,但不能阻止细胞正常生长,更不能诱发细胞凋亡.

A Cytotoxicity Study on Medical Wound Adhesive and Anastomotic Adhesive on Fibroblast L929 Cell of Mice
Abstract:Objective To investigate the indirect cytotoxicity of medical wound adhesive and anastomotic adhe-sive on fibroblast cells of mice. Methods The cytotoxicity of the two kinds of adhesives on fibroblast L929 cells of mice was detected. The effects of two kinds of medical adhesives on cell activity were detected using CCK-8 method;the effect on cell division and growth was detected using 4',6-diamidino-2-phenylindole ( DAPI) fluorescence staining method, and fluorescein isothiocyanate/propidium iodide ( FITC/PI) apoptosis staining method was used to detect the effect on apopto-sis. Results There was no significant difference in fibroblasts L929 activity of mice in the medical wound adhesive and anastomotic adhesive groups ( P>0. 05 ) , but the reduced values in the two groups were lower than that in the control group (P<0. 05). There were no significant differences in fluorescence staining cell count, total area proportion and sur-vival rate of normal cells among the three groups (P>0. 05). Conclusion The medical wound and anastomotic adhe-sives may significantly reduce the activity of peripheral adjacent cells with the similar reduction degree. However, they can not stop the normal of cells growth of cells and can not induce apoptosis.
