摘 要: | 目的 分析ECRG4基因在肝细胞肝癌组织(HCC)中的表达与甲基化情况,并探讨其临床意义.方法 分别采用免疫组化法、MSP-PCR两种方法,检测50例肝癌组织、对应癌旁组织及31例正常肝组织中的ECRG4蛋白的表达和ECRG4启动子区甲基化情况,并探讨两者之间的相关性.结果 ECRG4蛋白在肝癌组织、癌旁肝组织中表达水平显著低于正常肝组织(P<0.05),在肝癌组织中表达水平显著低于癌旁肝组织(P<0.05);且ECRG4肝癌组织的甲基化检出率明显高于癌旁肝组织及正常肝组织(P<0.05).ECRG4蛋白低表达率与ECRG4甲基化检出率明显相关(P均<0.05).结论 ECRG4在HCC组织中表达下调与其启动子区高甲基化存在明显的关联性,在肝癌的发生中发挥重要作用.

Expression of Esophageal Cancer Related Gene 4(ECRG4)in Hepatocellular Carcinoma Tissues and Its Clinical Significance |

Abstract: | Objective To study the expression of esophageal cancer-related gene 4(ECRG4)in hepatocellular carcinoma ( HCC) and the methylation condition of ECRG 4 of promoter region in gastric cancer tissues ,and their clinical significance.Meth-ods Immunohistochemistry and MSP-polymerase chain reaction ( PCR) were used to detect the protein expression level of ECRG 4 and its methylation condition in 50 cases of HCC tissues ,the adjacent non-tumor tissues and in 31 cases of normal liver tissue re-spectively.Explore the correlation between the protein expression of ECRG 4 and the methylation condition.Results The protein expression level of ECRG 4 in HCC tissues and adjacent non-tumor tissues both were significantly lower than normal liver tissues (P<0.05).The protein expression level of ECRG4 in HCC tissues was significantly lower than adjacent non-tumor tissues(P<0.05).The methylation rates of ECRG4 of promoter region in gastric cancer tissues were significantly higher than adjacent non -tumor tissues and normal gastric tissues (P<0.05).The low expression level and methylation rates of ECRG 4 had significantly positive correlation(P<0.05).Conclusion The down-regulation of ECRG4 expression is associated with high methylation of ECRG4 promoter region in gastric cancer tissues ,which is of great importance for the occurrence and development of HCC. |
