热处理对STZ诱导糖尿病性白内障大鼠晶状体上皮细胞生物学特性的影响

摘    要:目的 研究热处理对链脲佐菌素(streptozotoein,STZ)诱导的大鼠糖尿病性白内障的晶状体上皮细胞(lens epithelial cell,LEC)生物学特性的影响.方法 将30只SD大鼠随机分为3组A组为空白对照组,B组为STZ组,C组为热处理STZ组.每周用裂隙灯观察晶状体的变化,在实验6周后分别摘取眼球,取出晶状体.采用免疫组化技术检测热休克蛋白(heat shock protein,HSP)在各组LEC中的表达;运用考马斯亮兰与呈色法测定各组可溶性蛋白的浓度;呈色法测OD值,检测各组MDA、SOD及GSH的含量.结果 B、C组晶状体的混浊情况差异具有统计学意义(P<0.05),B组混浊程度明显高于C组,A、C组差异不显著.C组HSP70的表达高于B组,差异具有统计学意义(P<0.05);B组HSP70的表达高于A组,差异具有统计学意义(P<0.05).B组可溶性蛋白含量明显低于A、C组,差异有统计学意义(P<0.05),而A、C组差异不显著.A、B组晶状体MDA、SOD、GSH的含量均低于C组,差异具有统计学意义(P<0.05);A、B组之间差异不显著.结论 热处理可以提高HSP70的表达,并可能对STZ诱导糖尿病性白内障大鼠的晶状体上皮细胞具有保护作用.

Abstract:Objective To investigate the effect of thermotolerance on the lens epithelial cells(LECs)of Streptozotocin(STZ)-diabetic rats.Methods Thirty SD rats were randomly divided into three groups:Group A was set as normal control group;Group B as diabetic cataract group;Group C as diabetic cataract after thermotolerance treatment group.Lens were examined using a slitlamp once a week.At the end of 6th week after STZ injection,eyeballs were isolated and lens were excluded.Imunohistochemical method was used to detect the expression of HSP70 in each group.The content of water soluble protein and the levels of MDA,SOD,GSH were examined with streptavidin-peroxidases(S-P)method.Results LECs apoptosis of B group was significantly higher than that of C group(P<0.05),and there was no statistical difference between A group and C group.The expression of HSP70 of B group was great lower than that of C group(P<0.05),and higher than that of A group(P<0.05),and no statistical difference between A group and c group.The water soluble protein of B group was increased significantly than that of A group and that of C group(P<0.05),and no statistically different between A group and C group.MDA,SOD,GSH of B group and C group were lower than that of C group,respectively(P<0.05),no statistical difference between A group and C group.Conclusion The expression of HSP70 is significantly increased after thermotolerance treatment,wich may play a role in protecting lens epithelial cells.
