bcl-2和bax在口腔癌组织中表达与凋亡关系的研究

摘    要:目的 通过对口腔癌组织及癌旁正常组织中bcl 2和bax基因表达与本底凋亡细胞的研究 ,探讨口腔癌组织及癌旁正常组织中凋亡与基因表达的关系。方法 利用免疫组化法检测 67例口腔癌组织标本及 42例癌旁正常口腔组织中bcl 2和bax基因表达 ,利用TUNEL方法检测口腔癌组织标本中凋亡的情况。结果 bcl 2、bax基因在口腔癌组织及癌旁口腔组织中的阳性及阴性表达均具有显著性差异 ;两者在口腔癌组织中的阳性及阴性表达 ,其细胞凋亡的指数均有显著性差异。bcl 2、bax基因表达呈显著负相关。结论 口腔癌组织中bcl 2基因阳性表达时 ,其本底凋亡指数较阴性表达时低 ;而bax基因性表达时 ,其本底凋亡指数较阴性表达时高 ,两者在细胞凋亡过程中存在着拮抗作用的关系

Abstract:Objective To investigate the relationships between the exp re ssions of bcl-2 and bax genes and the spontaneous apoptosis in oral carcinomas. Methods 69 specimens of oral carcinoma and 42 specimens of n ormal oral cavity tissue were examined for the expressions of bcl-2 and bax gen es by immunohistochemistry, for apoptosis by in-situ TUNEL. The number of apopt otic cells was taken as the apoptotic index. Results The dete cted rates of bcl-2 and bax in carcinoma of oral cavity were 49.3% and 47.8% respectively, in oral cavity tissue, Those were 23.8% and 81.0% respectively. There were significant differences between the groups of positive negative expre ssion of bcl-2 or baxgene in the oral cavity carcinoma and oral cavity tissue a s well as between apoptotic indexes(AI) of cells of the groups of positive and n egative expression of bcl-2 or bax gene in the 69 specimens of oral cavity carc inoma. The AI of bcl-2 gene was significantly and negatively correlated to the AI of bax gene. Conclusions The AI of the groups of bax-posi tive expression are higher than those of the groups of bax-negative expression and the AI of the groups of bcl-2-positive expression are lower than those of the groups of bcl-2-negative.
