摘    要:目的观察二甲双胍对2型糖尿病模型大鼠脂肪组织中腺苷酸活化蛋白激酶α2(AMPKα2)的表达及对脂代谢的影响,探讨其改善血脂的可能机制。方法高脂饮食伴一次性腹腔注射链脲佐菌素的方法制备2型糖尿病模型,造模成功后随机分为模型组(DM-C)和治疗组(DM-T)。DM-T组给予盐酸二甲双胍灌胃治疗。测定大鼠治疗前后的体质量,实验末测定各组大鼠肾周及睾周脂肪质量,并检测各组大鼠总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)水平,计算大鼠脂肪质量/体质量比值。同时以半定量逆转录-聚合酶链反应法(RT-PCR)检测各组大鼠脂肪组织AMPKα2 mRNA的表达。结果与DM-C组比较,DM-T组大鼠脂肪组织AMPKα2的表达及血清HDL-C显著增高,AMPKα2(0.17±0.07)vs(1.06±0.50)(P〈0.01);HDL-C(0.44±0.08)mmol/L vs(0.88±0.12)mmol/L(P〈0.05)。DM-T组大鼠TC、TG和LDL-C比DM-C组显著降低,TC(5.22±1.70)mmol/L vs(1.87±0.57)mmol/L(P〈0.05);TG(5.11±0.92)mmol/L vs(0.78±0.39)mmol/L(P〈0.05);LDL-C(2.08±1.44)mmol/L vs(0.93±0.38)mmol/L(P〈0.05)。结论二甲双胍可能通过上调2型糖尿病大鼠脂肪组织AMPKα2表达,调节机体脂代谢。

Abstract:Objective To examine the effects of metformin on expression of AMPKα2,and lipid in adipose tissues of type 2 diabetic rats and investigate the possible mechanism of metformin improving lipid metabolism.Methods Type 2 dibetes models were prepared with high fat diets accompanied with intra-peritoneal injection of streptozotocin once and the model rats were divided into model group(DM-C) and treatment group(DM-T).DM-T group was treated with metformin by intragastric administration.The rat body mass before and after treatment was evaluated.The fat mass of adipose tissue around kindey and testicle was measured at last.Serum total cholesterol(TC),triglyceride(TG),high density lipoproteins cholesterol(HDL-C),low density lipoprotein cholesterol(LDL-C) were tested and adipose mass/body mass was calculated.The expression of AMPKα2 in adipose tissue was detected with RT-PCR.Results AMPKα2 MRNA expression,serum HDL in DM-T level were higher than those of DM-C,AMPKα2(0.17±0.07)vs(1.06±0.50)(P0.01);HDL(0.44±0.08) mmol/L vs(0.88±0.12) mmol/L(P0.05);serum TC,TG,LDL levels were lower in DM-T than in DM-C,TC(5.22±1.70) mmol/L vs(1.87±0.57) mmol/L(P0.05);TG(5.11±0.92)mmol/L vs(0.78±0.39) mmol/L(P0.05);LDL(2.08±1.44) mmol/L vs(0.93±0.38) mmol/L(P0.05).Conclusion Metformin may regulate lipid metabolism by up-regulating AMPKα2 expression in adipose tissues of type 2 diabetic rat.
