清肺口服液对Hep-2细胞感染呼吸道合胞病毒后IL-6、IL-8调控的研究

摘    要:目的探讨清肺口服液对人喉癌上皮细胞(Hep-2)感染呼吸道合胞病毒(RSV)后白细胞介素-6(IL-6)、白细胞介素-8(IL-8)表达的影响。方法分时段以清肺口服液含药血清、利巴韦林含药血清及空白血清干预RSV感染的体外培养的Hep-2细胞,48h及72h后收集各组细胞上清,采用酶联免疫吸附法(ELISA)测定各组人喉癌细胞IL-6、IL-8表达变化。结果 48h及72h后病毒模型组IL-6、IL-8的表达明显高于清肺口服液组(Ⅰ、Ⅱ、Ⅲ)、利巴韦林组(Ⅱ、Ⅲ),有显著差异,空白血清组(Ⅱ、Ⅲ)与病毒模型组无显著差异。清肺口服液组(Ⅰ、Ⅱ、Ⅲ)与利巴韦林组(Ⅱ、Ⅲ)比较,无显著差异。清肺口服液Ⅰ、Ⅱ、Ⅲ组之间比较,无显著差异;72h后各组IL-6、IL-8的表达与48h相比,无显著差异。结论清肺口服液可降低Hep-2感染RSV后IL-6、IL-8的过度表达,减轻炎症反应,提前给药更可获得良好的效果,这可能是清肺口服液抗病毒的机制之一。

Abstract:Objective:To investigate the effects of Qingfei Oral Liquid on expression of Interleukin(IL)-6 and Interleukin(IL)-8 in Human epithelial cells(Hep-2)infected with Respiratory Syncytial Virus(RSV).Methods:Hep-2 cells were intervened with serum containing Qingfei Oral Liquid,Ribavirin and blank serum respectively.In each of these 3 groups,intervention was conducted before,simultaneously or after RSV infection(Ⅰ,Ⅱ,Ⅲ).Expression levels of IL-6 and IL-8 were detected by enzyme linked immunosorbent assay(ELISA),after collecting the cellular supernatant.Results:After 48 and 72 hours of cultivation,levels of IL-6 and IL-8 in virus control group were significantly higher than those in Qingfei Oral Liquid groups(Ⅰ,Ⅱ,Ⅲ) or Ribavirin group(Ⅱ,Ⅲ).No significant differences were found between the blank serum control groups(Ⅱ,Ⅲ) and the virus control group.Difference between the Qingfei Oral Liquid groups(Ⅰ,Ⅱ,Ⅲ) and the Ribavirin groups(Ⅱ,Ⅲ) was insignificant either.Expressions of IL-6 and IL-8 within the Qingfei Oral Liquid group(Ⅰ,Ⅱ,Ⅲ) were slightly different after 48 or 72 hours of cultivation.Conclusion:Qingfei Oral Liquid can decrease the over-expression of IL-6 and IL-8 so as to relieve inflammation.Furthermore,better effects can be achieved when it is administrated in early stage of RSV infection.These foundings may relate to the antiviral mechanism of Qingfei Oral Liquid.
