海昆肾喜对阿霉素肾病大鼠肾组织PDGF-BB蛋白及mRNA表达的影响

摘    要:目的:研究海昆肾喜对阿霉素肾病大鼠肾组织血小板衍生生长因子-BB(PDGF-BB)蛋白及mRNA表达的影响。方法:将54只阿霉素肾病模型大鼠随机分为6组:正常组、假手术组、模型组、苯那普利组、海昆肾喜低剂量组及高剂量组。实验第10周观察各组大鼠24 h尿蛋白定量,肌酐(SCr)、尿素氮(BUN)水平及肾组织形态学变化;应用免疫组化及原位杂交方法分别观察PDGF-BB蛋白及mRNA在肾组织中的表达。结果:海昆肾喜低剂量组及高剂量组24 h尿蛋白定量和SCr、BUN水平较模型组均明显降低。PDGF-BB蛋白及mRNA的表达见于肾小管上皮细胞胞浆及肾小球系膜细胞,其表达较模型组显著减少。结论:海昆肾喜能抑制阿霉素肾病大鼠肾组织PDGF-BB蛋白及mRNA的表达,这可能是该药物防治肾小球硬化的作用机制之一。

Abstract:Objective: to investigate the effects of Haikun Shenxi on the expression of platelet-derived growth factor-BB(PDGF-BB) and mRNA in renal tissue of rat with adriamycin nephropathy.Methods: Rats model were established by unilateral nephrectomy and injecting adriamycin intraperitoneally,and the adriamycin-induced nephrotic rats were randomly divided into 6 groups: normal group,sham operation group,model group,Lotensin treatment group,Haikun Shenxi low and high dose treatment groups.10 weeks later,the 24 hours urine protein and blood biochemistry examinations and renal pathologic changes were observed,and the expression of PDGF-BB and mRNA were measured using immunohistochemical method.Results:compared with model group,proteinuria and the levels of serum creatinine(SCr),urea nitrogen(BUN) were decreased obviously in both Haikunshenxi low and high dose groups.The expression of PDGF-BB and mRNA were mostly presented in cytoplasm of renal tubular epithelial cells and mesangial area,and it could be reduced significantly after treatment(P<0.05).Con CluSion:the level of PDGF-BB and mRNA is high in renal tissue of adriamycin-induced nephrotic rats.This progress could be effectively inhibited by Haikunshenxi and the mechanism may be that it could control the excessive expression of PDGF-BB and mRNA.
