卡维地洛联合美托洛尔对大鼠心脏成纤维细胞白介素-6(IL-6)表达的影响

目的探讨联合应用两种不同作用机制的β受体阻滞剂与二者单独应用对大鼠心脏成纤维细胞白介素-6(IL-6)表达的影响。方法通过体外培养大鼠心脏成纤维细胞(CFs),应用不同剂量卡维地洛、美托洛尔以及两种联合干预细胞,MTT法检测细胞增殖,ELISA法检测IL-6的表达,并进行组间、组内对比研究。结果卡维地洛有抑制心脏成纤维细胞增殖的作用,卡维地洛和美托洛尔均对心脏成纤维细胞IL-6的表达有抑制作用,而且存在剂量依赖效应,卡维地洛与美托洛尔联用与单独使用它们相比无明显差异。结论卡维地洛与美托洛尔两药联合暂未发现存在协同作用。

Effects of combination of carvedilol and metoprolol to the expression of IL-6 of cardiac fibroblasts in rats

Objective To explore the effect of Carvedilol,Metoprolol,and their combination on the expression of IL-6 of cardiac fibroblasts.Methods Primary cardiac fibroblasts(CFs) were isolated by trypsin digestion method and identified by morphology and immunocytochemistry.Metoprolol or(and) Carvedilol with different concentrations were been used to intervention the growth of cells,proliferation and cell number weremeasured by 3-(4,5-Dimethylthiazol-2-yl)-2,5-diph-enyltetrazolium bromide(MTT).The enzyme-linked immunosorbent assay was employed to measure interleukin-6(IL-6)levels.Results The inhibition effect of different concentration Carvedilol on the cardiac fibroblasts proliferation was different.There was a dose-dependent relationship in the effect of Carvedilol inhibition in cardiac fibroblasts proliferation.Compared with the control group,the IL-6 levels in Carvedilol group was statistically different.Similarly,Metoprolol group was statistically different,but the combination group didn’t show any statistically difference contrasted Carvedilol or Metoprolol group.Conclusion The combination of Carvedilol or Metoprolol do not have any Combined Effect on the IL-6 levels of cardiac fibroblasts.
