丙泊酚的合成工艺改进

摘    要:目的合成丙泊酚。方法在浓硫酸存在下,以对羟基苯甲酸(1)为起始原料,与异丙醇反应,生成4-羟基-3,5-二异丙基苯甲酸(2),2再与氢氧化钠条件下加热脱羧生成目标化合物丙泊酚。结果及结论合成丙泊酚总收率大于64%(以1计),纯度大于99%。

Abstract:Objective To synthesize propofol.Methods 4-hydroxy-3,5-diisopropylbenzoic acid(2) was produced with p-hydroxy benzoic acid(1) and isopropanol in the conditionof concentrated sulfuric acid.Propofol was synthesis by decarboxylation of 4-hydroxy-3,5-diisopropyl benzoic acid(2) in the condition of high boiling point solvent and metal hydroxide in inert atmosphere.Results and Conclusion The overall yield rate of propofol synthesized was 64% and its purity was more than 99%.
