摘    要:目的:探讨应激反应对大鼠皮下移植瘤生长的影响。方法:将大鼠乳腺癌细胞Walker-256癌细胞株注入Wistar幼鼠腹腔,5d后抽取腹水,浓缩,经Wistar大鼠腹股沟皮下接种,建立Wistar大鼠皮下移植瘤模型。选取24只荷瘤大鼠,随机分为实验对照组(未烫伤)、轻度烫伤组(10%TBSA,100℃水中烫伤5s)、重度烫伤组(15%TBSA,100℃水中烫伤10s),每组8只。烫伤后12h尾静脉取血,ELISA法检测血自细胞介素-18(IL-113)水平,放射免疫法测定血清皮质醇水平;烫伤后1、7、13d测定皮下肿瘤体积,伤后13d进行肿瘤的病理学观察。结果:烫伤后大鼠血浆IL-1G水平较实验对照组(5.01±2.51)pg/m1]明显升高(P〈O.01),重度烫伤组(155.78±18.30)pg/ml]较轻度烫伤组(94.12±15.31)pg/m1]升高更加明显(P〈O.01);烫伤后大鼠血清皮质醇水平也较实验对照组(60.51±16.70)μg/L]明显升高(P〈O.01),重度烫伤组(115.78±17.42)μg/L]较轻度烫伤组(87.19±13.28)μg/L]升高更加明显(P〈O.05)。烫伤后1d,各组间肿瘤体积差异无统计学意义(P〉0.05);烫伤后7d和13d,对照组与轻度烫伤组肿瘤体积差异无显著性(P〉0.05),重度烫伤组肿瘤体积明显大于实验对照组和轻度烫伤组(P〈0.05)。病理学观察显示,烫伤组肿瘤细胞密度均较实验对照组大;各组均有纤维组织增生和炎细胞浸润,尤以重度烫伤组为甚。结论:重度烫伤后应激反应促进肿瘤的生长。

Abstract:Objective: To investigate the effects of stress on the growth of subcutaneously transplanted tumor after scald of different degrees in Wistar rats. Methods: Two Wistar immature rats were intraperitoneally injected Walker-256, the cells of breast cancer of rats. Ascites fluid was acquired 5 days later, and were centrifuged to ob- tain the ascites tumor cells. Subcutaneous tumor model in Wistar rats was reproduced .by subcutaneous implanta- tion of the tumor cells in groin. Twenty-four tumor bearing rats were randomly divided into control group (no scald) ,mild scald group scald at 100 ℃ for 5 seconds covering 10% total body surface area (TBSA)] ,and severe scald group (scald at 100 ℃ ,10 seconds covering 15% TBSA),with 8 rats in each group. Plasma interleukin-lβ (IL-1β) level was measured by enzyme-linked immunoadsorbent assay (ELISA) and serum cortisol level was meas- ured by radioimmunoassay 12 hours after scald injury. The size of tumor was observed on day 1,7,13, and tumor histopathology was examined on day 13. Results: IL-113 level in mild scald group (94.12±15.31) pg/ml)] was higher than that in control group (5.01± 2.51) pg/ml, P〈0.01 ]12 hours after scald, and it was significantly el- evated in severe scald group (155.78±18.30) pg/ml] compared with that of the mild scald group (P〈0.01). Serum cortisol levels in rats 12 hours after mild scald (87.19± 13.28) ug/L] were higher than that in control group (60.51 ± 16.70) ug/L, P〈0. 011, and it was significantly increased in severe scald group (115.78 ± 17.42)ug/L] compared with the mild scald group (P〈0.05). On day 1 after scald injury,there were no differ- ences among 3 groups in tumor volume (P〉0.05). On day 7 and 13 ,the size of tumor in the mild scald group was similar to that of the control group (P〈0.05), but it was larger in size in the severe scald group than that of the control group and mild scald group, the difference was significant (P〈0.05). Histopathological examination showed that more tumor cells were found in the two scald groups. Fibrous hyperplasia and infiltration of inflammatory cells were obvious in all the groups, especially in the severe scald group. Conclusions: Stress reaction produced by severe scald can accelerate the growth of tumor cells.
