| 摘 要： | 目的 检测 HL- 6 0 / ADM细胞化疗药物敏感性及 MRP表达。方法 以 HL- 6 0 / ADM细胞为实验对象 ,对化疗敏感的 HL - 6 0细胞为对照 ,MTT法检测药物敏感性 ,免疫细胞化学法检测 MRP蛋白的表达 ,PT- PCR法检测MRP m RNA表达。结果 两种细胞生长周期基本一致 ;HL - 6 0 / ADM细胞对 ADM、MTZ、VCR、H四种化疗药物 IC50 显著高于 HL- 6 0细胞 (P<0 .0 1) ,RF均在 2 0倍以上 ;HL- 6 0 / ADM表达 MRP m RNA,细胞膜表面 MRP阳性。结论 HL-6 0 / ADM细胞系适合体外 MDR研究。

| Abstract: | Objective To detect the multidrug resistance character of HL-60/ADM cell line.Methods HL-60/ADM cells with multidrug resistance induced by adriamycin (ADM) were included in the experimental group, and the sensitive HL-60 cells were used in the control guoup. The proliferation and the chemosensitivity of cells were studied by MTT assay method. The expression of multidrug resistance protein (MRP) on the membrane of cells was detected by immunocytochemistry. The expression of MRP mRNA was determined by semi-quantitive RT-PCR method.Result The IC 50 values, for ADM, MTZ, VCR and H, in HL-60/ADM cells were significantly higher than those in HL-60 cells (P<0.01), and the values of resistance fold were all more than 20 times. HL-60/ADM cells expressed abundnat MRP and MRP mRNA, but HL-60 cells did not express. Conclusion HL-60/ADM cell line can be used for MDR research in vitro. |
