高糖对大鼠肾系膜细胞增殖及NF-κBp65核转位的影响

摘    要:目的:探讨高糖对大鼠肾系膜细胞增殖及NF-κBp65核转位的影响。方法:大鼠HBZY-1肾系膜细胞进行体外培养,用5.6mmol/L葡萄糖(正常对照组)、10、20、30mmol/L高糖分别作用12、24、48小时,用噻唑蓝(MTT)法测各组肾系膜细胞增殖,western-blot法测各组肾系膜细胞NF-κBp65总蛋白的表达;双染免疫荧光激光共聚焦显微镜测各组肾系膜细胞NF-κBp65核转位。结果:48h内,各高糖组大鼠肾系膜细胞的增殖无统计学差异(P〉0.05);各高糖组NF-κBp65总蛋白的表达无统计学差异(P〉0.05);高糖组NF-κBp65核转位增加(P〈0.01),呈时间浓度依赖性。结论:在48h内,高糖直接促进肾系膜细胞NF-κBp65核转位导致NF-κB信号通路激活,并不促进大鼠肾系膜细胞的增殖及NF-κBp65总蛋白的表达增加。

Abstract:Objective:To explore the effect of high glucose on proliferation and nuclear translocation of NF-κBp65 in cultured rat glomerular mesangial cell(GMC).Methods:Cultured HBZY-1 rat GMCS were divided into normal control group(5.6mmol/L glucose) and 3 high glucose-treated groups(10,20,30mmol/L glucose).The proliferation of GMC was examined by MTT,the expression of NF-κBp65 protein was examined by western-blot;and the nuclear translocation of NF-κBp65 was observed by immunofluorescence and laser scanning confocal microscope respectively.Results:Within 48 hours,compared with normal control group,the proliferation and the expression of NF-κBp65 protein of GMC had no significant change in each high glucose-treated group(P0.05);the nuclear translocation of NF-κBp65 of GMC was increased obviously in 20,30mmol/L glucose(P0.01).Conclusions:High glucose can increase obviously the nuclear translocation of NF-κBp65 and activate the NF-κB signal pathway in cultured GMC,but not increase the proliferation of cell and the expression of NF-κBp65 protein of GMC.
