银染法观察大肠杆菌生物被膜变化初探

摘    要:目的探讨银染法下利用普通光学显微镜观察大肠杆菌生物被膜在药物作用前后的变化。方法采用腹膜透析液-腹膜透析管系统建立大肠杆菌生物被膜,分别用扫描电镜和银染法染色后应用普通光学显微镜来观察细菌生物被膜在药物作用前后的变化情况。结果使用银染法后用普通光镜观察大肠杆菌生物被膜变化的情况与用扫描电镜观察的结果一致。结论银染法下利用普通光学显微镜可以更为便捷地观察大肠杆菌生物膜的变化情况,且实验结果稳定、可靠。

Abstract:Objective To explore the changes of Escherichia coli biofilm in medicine effects observed by optical microscopy along with silver staining method.Methods Escherichia coli were cultivated in peritoneal dialysis solution-peritoneal dialysis cathete system to establish Escherichia coli biofilm model.After the biofilm was treated by medicine,we observe the changes of biofilm with optical microscope and scanning electronic microscope,respectively.Results After using silver staining method with the observation of optical microscope,the changes of Escherichia coli biofilm were showed no difference between optical microscope and scanning electronic microscope.Conclusions Through silver staining method,the changes of Escherichia coli biofilm can be more easily observed by optical microscope,and the experimental results were stable and reliable.
