维甲酸诱导的视网膜色素上皮细胞凋亡

摘    要:目的 研究维甲酸(rentinoic acid,RA)诱导的视网膜色素上皮(retinal pigment epithlium,RPE)细胞凋亡特征。 方法 体外培养的RPE细胞中加入10-510-610-7mol/L RA,应用吖啶橙荧光染色和核苷酸末端转移酶介导的dUIP末端标记法(Tdt-mediated dUIP nick end labelling method,TUNEL法)观察凋亡细胞。 结果 10-5、10-6、10-7mol/L RA诱导RPE细胞凋亡,凋亡细胞表现为细胞固缩,染色质聚.TUNEL法显示凋亡细胞DNA片断化.10-7、10-6、10-5mol/L RA作用5天时,凋亡指数分别为36.9、44.9%、61.4%。作用6天时,凋亡指数分别为48.0%、59.9%、74.2%。经统计学处理,同一RA浓度时,随作用时间延长凋亡指数增加;同一作用时间时,随RA浓度增大凋亡指数增加,差异有显著性(P<0.05). 结论 RA能诱导RPE细胞凋亡,且有很好的时间剂量效应。(中华眼底病杂志,1998,14:153-155)

Abstract:Purpose To investigate retinoic acid (RA) induced apoptosis in retinal pigment epithelial (RPE) cells.Methods10-5、10-610-7 mol/L were added to cultured PRE cells.Aridine orange fluorescence and TdT-mediated dUTP nick end labelling(TUNEL) techniques were used to observe apoptotic changes.Resultss10-5、10-6、10-7 mol/L RA induced apoptosis in RPE cells.Cell shringkage,chromatin condensation and nuclear DNA fragmentation of RPE cells were observed by TUNEL technique.When 10-7、10-6、10-5mol/L RA treated RPE cells for 5 days,apoptotic index(AI)was 36.9%、4409% and 61.4% respectively,and 48.0%、59.9%、74.2% for 6 days.At the same concentration of RA,AI increased when time prolonged.At the same day,AI increased when the concentration of RA rose.There was significant difference in the results(P<0.05).ConclusionOur results showed that RA-induced apoptosis in RPE cells was detected with a good dose and time response.(Chin J Ocul Fundus Dis,1998,14:153-155)
