葛花提取工艺研究

摘    要:目的:优选葛花的最佳提取工艺。方法:提取工艺研究采用正交试验法,筛选出葛花药材的最佳提Jgx-艺。采用HPLC法测定葛花有效成分鸢尾苷的含量。色谱条件:色谱柱:DiamosilC18(2)(150mm×4.6mm,5μm);柱温:室温;流动相:乙腈-水(20:80);波长:265nln;流速:1.0ml·min^-1;柱温:室温。结果:葛花的最佳提取工艺为药材加入40%乙醇溶液提取2次,第1次14倍量提取1.5h,第2次12倍量提取1.0h。鸢尾苷浓度在1.1~30μg·ml^-1。之间与峰面积线性关系良好,r=0.9999;鸢尾苷平均回收率为100.3%,RSD=2.0%。结论:本试验方法简单,专属性强,重复性好。

Abstract:To optimize the extracting condition of tectoridin in Flos Puerariae and determine the content. Method: The extracting process was optimized by orthogonal design. The content of tectoridin in Flos Puerariae was measured by HPLC with the following conditions:the chromatographic column was Diamosil Cls (2)( 150 mm× 4. 6 mm,5μm) with detection wavelength of 265 nm, the mobile phase was acetonitrile-water (20: g0) with the flow rate of 1.0ml·min^- 1, and the column temperature was ambient. Result: The optimal extrating conditions were the following:Flos Puerariae was recirculated with 40% ethonal for 2 times ,the first time was with 14-fold amount of Flos Puerariae for 1.5 h,and the second time with 12-fold amount of Flos Puerariae for 1.0 h. The average recovery of tectoridin was 100. 3% with RSD of 2. 0%. Conclusion: The studies show that the methods are sufficiently simple, sensitive and re- producible.
