环维黄杨星D对异丙肾上腺素致小鼠心肌肥厚保护作用的初步研究

摘    要:目的:初步研究环维黄杨星D(Cvb—D)对小鼠心肌肥厚的保护作用。方法:皮下注射异丙肾上腺素(Iso)1mg·kg^-1,bid,连续14d,制备小鼠心肌肥厚模型,各治疗组分别给予环维黄杨星D(6,9,12mg·kg^-1)和阳性对照卡托普利(100mg·kg^-1)灌胃给药,末次给药24h后处死小鼠,测量小鼠心脏质量指数(全心质量/体质量)和左心室质量指数(左心室质量/体质量),并观察心肌组织病理形态学改变,以评价Cvb—D对小鼠心肌肥厚的保护作用。结果:与空白对照组比较,Iso模型组小鼠心脏重量指数明显升高,病理切片可见心肌肥厚改变。与Iso组相比,Cvb—D高剂量组能明显降低小鼠心脏质量指数和左心室质量指数(P〈0.05或0.01),病理切片可见损害较模型组减轻。结论:环维黄杨星D对Iso诱导的小鼠心肌肥厚具有一定的保护作用,其机制有待进一步研究阐明。

Abstract:To study the protective effect of cyclovirobuxinum D (Cvb-D) on cardiac hypertrophy induced by isoprenaline (Iso) in mice. Method: Mice cardiac hypertrophy model was established by administration of isoproterenol (Iso) 1 mg·kg^-1 sc, twice daily for 14 days. The therapy groups were intragastric administration of Cvb-D (6,9 and 12 mg·kg^-1) or captopril (100 mg· kg^-1 ). The heart weight index (heart weight/body weight, HW/BW), left ventricular weight index (left ventricular weight/body weight, LVW/BW) and myocardial morphological changes were determined to evaluate the protective effect of Cvb-D on cardiac hypertrophy. Result: Compared with those of control group, the HW/BW and LVW/BW of Iso group obviously increased, and the pathologic changes could be seen. Cvb-D ( 12 mg·kg^-1 ) could obviously decrease the heart indices in mice (P 〈0. 05 or P 〈0. O1 ) ,and also improve the pathologic damages of cardiac hypertrophy. Conclusion : Cvb-D can inhibit cardiac hypertrophy induced by Iso, and the mech anism needs to be further studied.
