清醒Wistar大鼠缺氧性肺动脉高压模型的建立

摘    要:为排除麻醉药物对大鼠肺血管反应性的影响,建立了Wistar大鼠清醒模型肺循环的研宄方法,并比较在麻醉及清醒状态下大鼠缺氧性肺动脉压升高反应的差异。结果表明:麻醉状态大鼠肺动脉压对缺氧刺激的反应不明显,清醒大鼠肺动脉压可因缺氧刺激明显升高。清醒大鼠的模型能比较真实地反映缺氧刺激引起肺循环机能状态的改变,麻醉药物的影响是肺循环研究申应当注意的问题。

Abstract:Pentobarbital is commonly used as an anesthetic in animal experiments. We had reported that pentobarbital inhibited pulmonary vascular reactivity, so we tried to establish a model of acute hypoxic pulmonary hypertension in awake rats to avoid the inhibition of pentobarbital on pulmonary vasculature. This paper introduced the method to establish an awake rat model for pulmonary vascular circulation research and also showed the comparison of acute hypoxic responses both in anesthetic rats and in awake ones.In anesthetic rats, hypoxic challenge caused small increase of pulmonary artery pressure, yet, the same stimulation produced severe pulmonary hypertension in awake rats. These results suggested that it is important to use the awake rat model for the study of hypoxic pulmonary vascular responses in rats.
