口腔鳞癌组织中人乳头瘤病毒16型的原位杂交研究

摘    要:目的:探讨HPV16-E6基因在口腔粘膜白斑,口腔鳞状细胞癌组织中的表达及其在口腔鳞癌发生、发展过程中的意义及分布特征。方法:应用地高辛标记的HPV16-E6的寡核酸探针分别对口腔鳞状细胞癌组织,口腔粘膜白斑组织,宫颈癌组织及正常组织中进行原位杂交,以检测各组织中HPV16型的DNA表达和分布情况,结果:口腔鳞状细胞癌组织HPV16阳性率46.67%;口腔粘膜白斑组织阳性率60%。HPV16E6基因阳性信号集中分布于癌细胞胞核中。结论:原位杂交方法可用来检测OSCC组织中HPV16型DNA的存在并能准确组织定位;较高的阳性率进一步支持高危型HPV16型的感染与口腔鳞状细胞癌的发生有关;在癌前病损组织中的高危型HPV16的检m率高于口腔鳞癌组织,说明HPV在口腔鳞癌发生的早期阶段可能具有致癌作用。

Abstract:Objective:To detect the expression of HPV16 E6 on oral leukoplakia and oral squamous cell carcinoma and explore its significance during the development and progression of oral squamous cell carcinoma and its characteristic of distribution. Methods: Using digoxin labelled human papillomavirus (HPV) 16 probes and hybridization in situ technique to detect expression of HPV16 E6 gene in OSCC and OLk tissues and its positive signal distribution on these tissues. Results: The results showed that the positive ratio was 46.67% in OSCC, 60% in begin oral lesions and100% in cervix cancer, no case of normal tissues was positive. Those positive signal distribute in nucleolus. Conclusions: It was suggested that hybridization in situ technique can detect HPV16 E6 gene and precisely location in nucleolus;In the same time,HPV16 E6 gene maybe closely related with the cause of OSCC especially in early development of OSSC.
