摘    要:目的探讨三七总皂甙对大鼠肝移植I/R损伤的影响及可能机制。方法SD大鼠随机分为3组:假手术(SO)组,生理盐水灌注(NS)组,三七总皂甙灌注(PNS)组。于供肝再灌注后2h处死动物取出肝脏标本,检测肝细胞凋亡及BCL-2,MDA水平。结果假手术组肝细胞凋亡少见,NS组与PNS组与SO组比较,肝细胞凋亡显著增高。PNS组与NS组比较,肝细胞凋亡显著降低。肝细胞BCL-2蛋白的阳性表达,PNS组与NS组和SO组比,差异有显著性。MDA含量:PNS组与NS组和SO组比,差异有显著性。结论PNS预处理对肝脏缺血/再灌注损伤(I/R)具有保护作用,其可能机制为抑制氧自由基生成,上调调控基因BCL-2蛋白的表达抑制肝细胞凋亡。

Abstract:To investigate the effect of Panax notoginseng on Ischemia-reperfusion injury in liver transplantation and the mechanisms. SD rats were randomly divided into sham operation(SO) group ,NS infusion group and Panax notoginseng infusion group. 2 hours after reperfusion, the rats were killed and the liver tissue was sampled to observe the apoptosis and expression of bcl-2, bax and MDA on hepatocyte. Compared with SO group in which nearly no apoptosis was found, there was significant difference of the apoptosis between NS group and PNS group. At the same time, the apoptosis decreased greatly in PNS group Compared with NS group. Positive rate of bcl-2 protein on the hepatocyte: There was a significant difference in PNS group while Compared with SO group and NS group. The content of MDA: There was significant difference in PNS group while Compared with SO group and NS group. Conclusions] Panax notoginseng pretreatment can provide effective protection against liver Ischemia-reperfusion injury. The mechanisms may be to depress the production of oxygen free radical and elevate the expression of bcl-2 protein on hepatocyte.
