大豆异黄酮配伍叶酸对神经管畸形胎鼠神经细胞中凋亡基因表达的影响

摘    要:目的 :观察大豆异黄酮和叶酸的联合干预对神经管畸形胎鼠神经细胞凋亡基因表达情况的影响 ,并研究其干预机制。方法 :将大鼠孕鼠随机分成对照组、模型组、单独干预组和配伍干预组后进行不同的处理。孕 1 4 d时处死孕鼠取出胎鼠 ,将胎鼠的脑组织进行固定和包埋后进行免疫组化染色 ,在高倍镜下进行观察。结果 :环磷酰胺可以导致凋亡基因 p5 3和 bax表达的增强 ,而大豆异黄酮和叶酸的干预可以增强凋亡基因 Bcl- 2的表达而减弱 p5 3和 Bax的表达 ,且联合干预的效果较好。结论 :3种凋亡基因的表达可能联合参与了神经管畸形的发生机制 ,大豆异黄酮可能通过对神经管畸形胎鼠神经细胞凋亡基因表达的影响而增强叶酸对神经管畸形的干预作用。

Abstract:Objective:To observe the effect of isoflavone cooperating with folic acid on the expression of apoptotic genes in neurons of Neural Tube Defects rat embryos induced by cyclophosphamide and to investigate the mechanism.Methods:The pregnant rats were randomly divided into control group,model group,solo-administration group and co-administration group. On the 14th day of gestation, the brains of the embryos were fixed in the neutral formalin solution and made to paraffin sections for immuohistochemmistry staining.Results:Cyclophosphamide can induce the expression of apoptotic genes p53 and bax increased, while the administration of isoflavone and folic acid can intensify the expression of bcl-2 and lessen the expression of p53 and bax. The effect of co-administration was better.Conclusion:The expression of the three apoptotic genes might be involved in the mechanism of Neural Tube Defects; Isoflavone might effect the expression of aopoptotic genes to reinforce the protective effect of folic acid on Neural Tube Defects .
