压力负荷性左室肥厚大鼠心肌钠钙交换体和肌浆网钙泵的表达

目的 观察压力负荷性左室肥厚大鼠心功能异常及心肌钠钙交换体(NCX)和肌浆网钙泵(SERCA2a)的表达变化.方法 缩窄大鼠腹主动脉制备压力负荷性心肌肥厚模型,测定在体血流动力学及左室重量指数(LVWI),用RT-PCR和Western blot法检测左室组织NCX及SERCA2a的表达.结果 与假手术组相比,模型大鼠左室收缩压(LVSP)及左室舒张末压(LVEDP)均显著升高(P<0.01,P<0.001);左室重量指数显著增加(P<0.001)及左室NCX mRNA表达上调(P

NCX and SERCA2 a expression in myocardial tissues in rats with pressure overload-induced left ventricular hypertrophy

Objective To observe the abnormity of heart function in rats with pressure overload-induced left ventricular hypertrophy and the changes of NCX,SERCA2a expression in myocardial tissues. Methods Cardiac hypertrophy was induced by clipping the abdominal aorta in rats. The cardiac hypertrophy was evaluated by Left ventricular weight index(LVWI,left ventricular weight/body weight). NCX, SERCA2a mRNA and protein expressions in left ventricular tissues were determined by half-quantitative RT-PCR and Western blot normalized to abundance of GAPDH mRNA and protein,respectively. Results LVSP and LVEDP were obviously enhanced(P<0.01,P<0.001) in rats with cardiac hypertrophy in vivo. LVWI was increased remarkably(P<0.001)in model group. RT-PCR and Western blot results demonstrated that NCX mRNA expression was upregulated(P<0.05),and SERCA2a mRNA and protein expression were downregulated(P<0.05) significantly in model group as compared with sham group. Conclusion Abnormity of heart function,especially diastolic function was observed. in rats with pressure overload-induced cardiac hypertrophy,and that may be the outcome of SERCA2a expressions in left down-regulation ventricular tissues.
