大剂量头孢唑啉耳毒性的实验研究

摘    要:目的观察大剂量头孢唑啉在铁缺乏条件下对新西兰大白兔耳蜗毛细胞的影响。方法60只健康新西兰大白兔,随机分成A、B、C、D组,每组15只。A、B两组喂标准饲料4周,C、D两组喂缺铁饲料4周,后两组制成铁缺乏动物模型。B、D两组经耳静脉注射头孢唑啉(0.5g/kg,2次/d,共14天),A、C两组经耳静脉注射等体积生理盐水做对照,停药后第15天处死作耳蜗扫描电镜及激光共聚焦显微镜观察。结果缺铁头孢唑啉组(D组)停药后第15天,有9个耳蜗标本(60%)存在不同程度内、外毛细胞静纤毛散乱、扭曲、缺失,以中区、顶区损害较重。结论大剂量头孢唑啉在铁缺乏条件下可造成新西兰大白兔耳蜗内、外毛细胞静纤毛不同程度损伤。

Abstract:Objective To observe the changs of cochlear hair cell induced by large dose cephazoline in iron - deficient New Zealand white rabbit. Methods 60 healthy New Zealand rabbit were randomly divided into 4 groups. The animal of group A,B were fed with standard diet and group C,D were given the basic iron - deficent diet for 4weeks. The rats of groups B, D were received intravenous injection of large dose cephazoline for 14 days. The ehangs of cochlear hair cell were observed by scanning electron microscope at 15days after injection. Results The scanning electron microscope showed stereocilia of the outer and inner hairy cell were disarrangment and some disappeared at middle, apical and base cochlear area. Conclusion The results suggest that large dose cephazoline may have potential effect on the function and structure of cochlear hairy cell when iron deficiency exists.
