冰冻红细胞制备方法的改进

目的建立1种冰冻红细胞最佳制备方法,用于稀有血型血液的长期保存。方法以甘油为保护剂,低温保存红细胞,使用前水浴解冻,用不同浓度的洗涤液洗涤去除甘油,以红细胞回收率、游离血红蛋白含量、残余甘油量、体外溶血试验和无菌试验为质量控制项目。结果储存6 d内的全血或悬浮红细胞,10 mL/min、150次/min水平摇摆条件下分2次加入57.1%的甘油溶液,中间静置平衡10 min,使甘油浓度达到40%(w/v),-80℃保存。数周后,42℃水浴解冻,15 mL/min、150次/min水平摇摆条件下分别用9%NaCl溶液、羟乙基淀粉40氯化钠溶液、0.9%NaCl溶液洗涤去除甘油,再用0.9%NaCl溶液混悬。18袋制品各项质量指标好于原传统方法,且均符合国家要求,临床使用效果良好。结论此法优于传统方法,可在实际中应用。

Improvement on the method in making the frozen red blood cells

Purpose To establish a best method for making the frozen red blood cells,and to perserve the blood for rare blood group for a long-term.Methods Use glycerine as protective fluid,keep RBC in a low temperature,unfreeze it with water before using,and remove the glycerine with several different washing solutions in concentration.The rate of recovery of RBC,the content of free Hb,the content of the glycerine residue,the test of external hemolysis and the aseptic test are taken as the projects of quality control.Results The whole blood or red cells suspension stored in 6 days,was added into 57.1% glycerine solution for 2 times in 10 ml/min and 150 bpm zero-level swing condition,during keeping untouched for 10 mins,to make the glycerine concentration up to 40%(w/v),which was kept in the refrigerator at-80℃.Several weeks later,unfreeze it with 42℃ water,and in 15 ml/min and 150 bpm zero-level swing condition,removed glycerine using 9% NaCl solution,hydroxyethyl starch 40 sodium chloride solution,and 0.9% NaCl solution,and then mix-suspended it with 0.9% NaCl solution. All the quality-indexes of 18 bags of production prior to the traditional methods,which comply with to the state demands,also acguired a good clinical efficiency.Conclusion The method is better than the traditional methods,and can be used in practical application.
