摘    要:目的:检测大鼠脊髓损伤后神经细胞的DNA损伤、凋亡及P53蛋白的表达。方法:采用大鼠脊髓急性压迫损伤模型,应用原位末端标记、DNA凝胶电泳、免疫组化等方法,对大鼠损伤脊髓进行检测。结果:脊髓损伤后4h开始出现凋亡细胞及P53蛋白表达,并在24h达到高峰。两者阳性细胞形态及分布有相似之处。DNA电泳可见24h组出现特征性条带。结论:大鼠脊髓损伤后出现大量的DNA损伤和神经细胞凋亡。P53蛋白可能参与了诱导神经细胞凋亡的过程

Abstract:Objective: To determine the DNA damage, neuronal cells apoptosis and the expression of P53 protein in the rat spinal cord after compression injury. Methods: We used terminal deoxynucleotidyl transferase mediated dUPT nick end labeling (TUNEL), DNA gel electrophoresis and immunohistochemistry techniques to detect DNA fragmentation in the injured rat spinal cord. Results: The apoptosic cells and P53 protein presented at 4 h after spinal cord injury, with a maximum presence at 24 h. DNA fragmentation presented typical ladder pattern on agarose gel at 24 h. Conclusion: There are lots of neuronal cells apoptosis and DNA damage in the spinal cord after injury. The P53 protein may be play an important role in induction of neuronal cells to apoptosis.
