Lazaroid对早期糖尿病大鼠视网膜的保护作用

摘    要:目的探讨Lazaroid对早期糖尿病大鼠视网膜的保护作用。方法20只SD大鼠采用链脲佐菌素(STZ)建立糖尿病动物模型,10只于成模2周后经尾静脉注射Lazaroid进行干预(糖尿病Lazaroid干预组),其余10只不予干预(糖尿病不干预组)。另设立不建模且不干预的空白对照组(n=10)。分别于实验前后进行视网膜电流图检查,对视网膜组织切片进行透射电子显微镜观察。结果糖尿病Lazaroid干预组和空白对照组在实验前后的暗适应最大电反应b波、振荡电位(OPs)振幅值差异无统计意义(P>0.05)。糖尿病不干预组实验前后的暗适应最大电反应b波、OPs振幅值明显下降(P<0.05);与空白对照组比较,成模4周后的糖尿病不干预组大鼠的神经节细胞内出现线粒体的改变;而同期糖尿病Lazaroid干预组大鼠视网膜未出现明显神经节细胞的改变。结论糖尿病动物模型在建立的早期已经出现视觉电生理的变化,并且有神经细胞超微结构的改变;全身使用Lazaroid对早期糖尿病大鼠模型视网膜神经元具有保护作用。

Abstract:Objective To investigate the protective effects of Lazaroid on early-stage diabetic retinopathy of rats.Methods Diabetic animal models were established by streptozotocin(STZ)in 20 rats.Ten rats were treated by intravenous injection of Lazaroid 2 weeks after model establishment(Lazaroid-treated diabetic group),and the other 10 received no intervention(STZ-induced diabetic group).Another 10 rats without STZ-induced diabetes and interventions were served as controls(control group).Electroretinogram examinations were performed before and after experiment,and retinal sections were observed by transmission electron microscopy. Results There was no significant difference in the amplitudes of oscillatory potentials(OPs) and b wave between Lazaroid-treated diabetic group and control group before and after experiment(P>0.05).Amplitudes of OPs and b wave were significantly decreased after experiment in STZinduced diabetic group(P<0.05).Compared with control group,mitochondrial metamorphosis in ganglion cells occurred in STZ-induced diabetic group 4 weeks after model establishment,whereas no obvious mitochondrial metamorphosis was observednd in Lazaroid-treated diabetic group. Conclusion Visual electrophysiological changes take place in the earlier stage of diabetic animal model with neurocyte ultrastructural changes.Lazaroid has protective effects on diabetic retinopathy of rats.
