摘    要:目的建立珠子参多糖的含量测定方法,并比较不同产地珠子参中多糖含量的差异。方法采用水提醇沉法对珠子参中多糖进行提取、纯化,采用蒽酮-硫酸比色法对不同产地珠子参中多糖进行含量测定,并对测定结果进行聚类分析。结果在0.0057 mg/mL~0.0190 mg/mL葡萄糖浓度C与吸光度A线性关系良好(r=0.9998),平均回收率为101.55%,RSD 2.2%,不同产地的珠子参多糖含量在3.26%~9.36%,不同产地珠子参多糖存在差异。结论本试验建立的珠子参中多糖含量测定方法简便、稳定、可行,不同产地珠子参中多糖含量的差异较大,该结果为珠子参中多糖质量控制和相关产品的开发利用提供了理论依据。

Abstract:Objective To establish a method for the determination of polysaccharides in Rhizoma panacis majoris,and prepare the difference of content from different producing areas. Methods Polysaccharides was extracted and purified from Rhizoma panacis majoris by water extraction and alcohol precipitation,and the content of polysaccharides was determined by anthrone- sulphuric acid colorimetry,then develop the method of hierarchical cluster analysis by using the results. Result The glucose concentration value C and absorbance A were linear in the range of 0. 0057 mg /mL ~ 0. 0190 mg /mL( r =0. 9998),The average recovery was 101. 55%,RSD2. 2%. The content of polysaccharides in Rhizoma panacis majoris from various producing areas varied from 3. 26% ~ 9. 36%. The amounts of polysaccharides from various habitats are different. Conclusion The method for determinating polysaccharides was simple,stable and workable. polysaccharides content was significantly different from different producing areas,the results provided references for polysaccharides content quality control and developing and using it.
