碘化钾对人体皮肤成纤维细胞增殖活性的影响

摘    要:目的探讨不同浓度的碘化钾(KI)对人体皮肤成纤维细胞(HSF)增殖活性的影响。方法将体外培养的HSF分成7组含不同浓度的KI(0、1?000、3?000、5?000、7?000、9?000、11?000?μg/L),每组含8个平行样本。完全培养基分别培养24?h后,观察HSF形态变化,用流式细胞术和酶联免疫吸附试验(ELISA)方法检测HSF增殖活性。结果KI的浓度在1?000、3?000、5?000?μg/L时成纤维的增殖活性随着碘浓度的增加而升高(P<0.05);5?000、7?000?μg/L组细胞增殖活性最高,两组间增殖活性差异无统计学意义(P>0.05);7?000、9?000、11?000?μg/L?时HSF增殖活性随着KI浓度的增加而降低(P<0.05)。结论KI浓度在5?000?μg/L以下会促进HSF增殖活性,高于7?000?μg/L时反而会抑制HSF增殖活性。

Abstract:To study the effects of KI on proliferative activity of human skin fibroblasts at different doses. MethodsHuman skin fibroblasts (HSF) were treated with KI 0, 1?000,3?000,5?000,7?000,9?000, and 11?000?μg/L for 24?h and to observe the morphology of fibroblasts. The methods of FCM and ELISA were used to detect the proliferation activity of human skin fibroblasts. ResultsKI at a certain range of concentration(<5?000?μg/L)could significantly increase proliferative activity of human skin fibroblasts but decreases the proliferative activity at a certain range of concentration(>7?000?μg/L). The proliferative activity was the highest from 5?000?μg/L to 7?000?μg/L, but there were no significant differences between them. ConclusionKI with an exposure level<5?000?μg/L can increase the proliferative activity of human skin fibroblasts, whereas it decrease the activity when its exposure >7?000?μg/L.
