化痰散结方对人肺癌细胞Fas-FasL和TNFR1信号通路的影响

摘    要:目的观察化痰散结方含药血清,对人肺癌 SPC-A1细胞 Fas-FasL 和 TNFR1的表达的影响。方法通过血清药理学方法,选择不同浓度的含化痰散结方的兔血清,与人肺癌细胞 SPC-A1共同孵育不同时间后,应用倒置相差显微镜对细胞生长和凋亡状况进行形态学观察,应用 SABC 法检测 SPC-A1细胞 Fas 和FasL 的表达。用原位杂交方法检测 SPC-A1细胞 TNFR1的表达。结果人肺癌细胞经含化痰散结方的兔血清作用后,由贴壁生长而逐渐脱落,外形变圆,体积缩小,折光性增加;SABC 法显示,化痰散结方含药血清可上调 SPC-A1细胞 Fas(P<0.05),而且可下调 SPC-A1细胞 FasL 的表达(P<0.05)。原位杂交方法显示,含药血清组 SPc-A1细胞 TNFR1的表达明显高于不含药血清组(P<0.01)。结论化痰散结方含药血清可诱导人肺癌细胞凋亡,其机制可能与上调 SPC-A1细胞 Fas、下调其 FasL 以及上调 TNFR1有关。

Effect of Removing Phlegm and Resolving Masses Recipe on Signal Pathway of Fas,FasL and TNFR1 of SPC-A1 Cell in Lung Cancer

Abstract:Objective To observe the effect of Removing Phlegm and Resolving Masses Recipe(RPRMR)contained pharma- ceutic serum on the expression of Fas-FasL and tumor necrosis factor R1(TNFR1)of SPC-Al cell in lung cancer.Meth- ods Different concentrations of RPRMR contained pharmaceutic rabbit serum were chosen by serum pharmacological method.After co-incubated with SPC-Al cell of human lung cancer at different time points,morphological observation of the growth and apoptosis of cells was performed by inverted phase microscope.Fas and FasL expression of SPC-Al cells were determined by SABC method,and TNFR1 expression of SPC-Al cells determined by in situ hybridization.Results After acted by RPRMR contained rabbit serum,human lung cancer cells changed from growing against wall to peeling off gradually,with rounding shape,shrinking volume,and increased refractivity.SABC method showed that RPRMR contained serum could up-regulate Fas expression and down-regulate FasL expression of SPC-Al(P<0.05).In situ hybridization method showed that TNFR1 expression was higher in SPC-Al contained serum than in the control group(P<0.01). Conclusion RPRMR contained serum could induce the cell apoptosis of human lung cancer,whose mechanism might be related to the up-regulation of Fas expression and down-regulation of FasL expression of SPC-Al cell,as well as up-regu- lation of TNFR1.
