摘    要:目的 研究中医体质与免疫共刺激分子单核苷酸多态性之间的相关性,探究中医体质分类理论与机体免疫系统功能间联系.方法 利用标准化中医体质量表结合中医四诊综合辨识,对359例志愿者进行中医体质辨识,利用聚合酶链式反应限制片段长度多态性(PCR-RFLP)方法鉴定其免疫共刺激分子CD40、CD28基因主要标签单核苷酸多态性位点(tagSNP)基因型.结果 对359例志愿者进行中医体质分类,发现CD40基因rs1883832位点基因型和等位基因在平和质和阳虚质中的分布频率存在统计学差异(P <0.05);CD28基因单体型GGCCTTCATAA与ACCTTTCATGA在平和质、气虚质和阳虚质三种中医体质中分布频率存在统计学差异(P<0.05).结论 中医体质论与免疫共刺激分子基因单核苷酸多态性存在关联性,免疫共刺激分子基因单核苷酸多态性所造成的免疫功能个体差异可能是不同中医体质个体存在疾病易感性差异的理论依据之一.

Association between types of traditional Chinese medicine constitutions and polymorphisms of immune co-stimulatory molecule genes
Abstract:Objective To investigate the association between types of traditional Chinese medicine (TCM) constitutions and polymorphisms of immune co-stimulatory molecule genes and to explore the relationship between the theory of constitution of TCM and immune system functions.Methods 359 subjects were enrolled in this study,whose constitutions of TCM were identified according to the standardized constitutional questionnaire and TCM four diagnostic information.CD40 and CD28 tagSNPs were genotyped by PCR-RFLP assay.Results Of all the 359 subjects,Normality (n =105,29.2%),Yang-deficiency (n =67,18.7%),Yin-deficiency (n =41,11.4%),Qi-deficiency (n =36,10.0%),Wetness-heat (n =19,5.3%),Depressed (n =40,11.1%),Phlegm-wetness (n =21,5.8%),Blood stasis (n =17,4.7 %),and inherited special constitution (n =13,3.6%) were determined.We found that the distribution of CD40 rs1883832 genotypes was significantly different among TCM constitutions.The frequencies of haplotypes GGCCTTCATAA and ACCTTTCATGA in CD28 gene were significantly different among TCM constitutions.Conclusion Polymorphisms of immune costimulatory molecuLe gene appear to be associated with TCM constitutions.Immune function difference resuLting from co-stimulatory molecule genes polymorphisms may influence disease susceptibility among different TCM constitutions.
