摘    要:目的:探讨脂联素基因3’非编码区(UTR)3224C/T多态性与中国上海地区男性高血压患者发生糖代谢异常的关系。方法:研究对象共729例,其中男性高血压伴糖耐量异常(IGT)患者359例,男性高血压伴糖耐量正常(NGT)患者370例。采用基质辅助激光解吸附电离飞行时间质谱(MALDI-TOF)检测技术鉴定脂联素基因3224C/T多态性。同步进行口服葡萄糖耐量试验(OGTT)及胰岛素释放实验。结果:脂联素基因3224C/T多态等位基因频率和基因型分布,在IGT和NGT2组间分布有显著差异(均P

Abstract:Objective To investigate the adiponectin gene 3224C/T polymorphism, and its relationship with abnormal glucose metabolism in male hypertensives. Methods 3224C/T genotype of adiponectin gene in 359 male essential hypertensives concomitant with impaired glucose tolerance (IGT) and 370 male essential hypertensives with normal glucose tolerance (NGT) was determined by matrix,assisted laser desorption/ionization time,of,flight mass spectrometry (MALDI,TOF) assay. Simultaneously, OGTT and insulin release test (InRT) were conducted. Results Significant differences for allele and genotype distribution of 3224 C/T were observed between IGT and NGT group (P<0.05), the T allele carriers demonstrated higher risk of IGT than C allele carriers, with odds ratio(OR) of 1.35 (95% CI, 1.06-1.71). The patients with homozygous TT genotype demonstrated higher plasma glucose level at OGTT 30 min, first, and second,phase of insulin release than those with CC and CT genotypes (P<0.05). Conclusions Adiponectin gene 3224C/T polymorphism is associated with abnormal glucose metabolism in male essential hypertensives.
