IgGimmunoreactivities(IgGIR) in the rat central nervous system following administration of the antineurons antibody

Abstract:Using porcine spinal motoneurons(Mns) an immunogen to Balb/c mice, we got monoclonal antibody (McAb,36F6) against Mns.McAb(IgG) was able to crossreact with human and rat's Mns.36F6 enters Mns by axoplasmic transport retrogradely to the their perikarya, but not enters the primary sensory neurons,when the McAb was injected to muscles.Modified ABC method was used to localized IgGimmunoreactivity (IgGIR) of the mouse IgG(36F6) in rat central nervous system.With a survival time 4872h after im injection of (36F6),a obvious transcytosis of McAb occurred from retrograde labeled Mns to interneurons in the lamina , at the level of spinal cord and even the preMns of the reticular formation (RF) in the brain stem.The pericellular staining neurons of IgGIR and the intracellular staining neurons of IgGIR in RF occurred in the same section,suggested the transcytosis from spinoreticular fibers to reticular neurons.IgGIR cells were also observed in the hypothalamic paraventricular nucleus,supraoptic nocleus,neocortical pyramidal cells,ependymal cells and infrequently or randomly in perivascular glia cells and/or neurons.
