Lewis全细胞肿瘤疫苗防治C57BL/6小鼠Lewis肺癌的实验研究

摘    要:目的探讨Lewis全细胞肿瘤疫苗对小鼠Lewis肺癌的防治作用。方法用Lewis全肿瘤疫苗免疫C57小鼠4次,其中首次免疫用含弗氏佐剂油剂疫苗,第4次免疫的同时在小鼠右腋下接种Lewis肺癌细胞,定期观察小鼠成瘤情况及小鼠免疫状态变化。用流式细胞术检测疫苗组和对照组小鼠外周血CD4 T细胞,CD8 T细胞及CD19 B细胞百分比,以及CD4/CD8的变化。结果疫苗组与对照组小鼠成瘤率、成瘤时间、肿瘤大小差异均无显著性;疫苗组与对照组相比,其外周血CD4 、CD8 T淋巴细胞亚群的百分比差异无显著性,CD4/CD8比值的差异也无显著性,疫苗组外周血CD19 B淋巴细胞百分比略高于对照组,差异有显著性(P=0.014)。结论用Lewis肿瘤全细胞疫苗不能提高C57BL/6小鼠的T淋巴细胞免疫功能,可以提高其B淋巴细胞免疫,但不能使小鼠获得有效的免疫保护。

Abstract:Objective To observe the tumor formation and the efficacy of immune response of C57BL/6 mice after the mice were inoculated an autologous tumor-cell vaccine against Lewis lung carcinoma.Methods Two groups of mice(vaccine group and control group) were inoculated,totally 4 times.When the third vaccine was inoculated,the mice were injected with Lewis lung tumor tissue suspension at right axillary subcutaneously region at the same time.Then tumor formation was observed.The subsets of the lymphocytes(CD4 -T cell,CD8 -T cell and CD19 B cell) in peripheral blood of the two groups of mice were also assayed with flow cytometry.Results Two groups of mice have the same rate of tumor-formation(100%).And there is no significant difference in time of tumor-formation and tumor volume between the two groups.Contrasted with the control group,the vaccine group shows no significant difference in the subsets of the lymphocytes in peripheral blood.Conclusion This study indicates that the Lewis autologous tumor-cell vaccine can enhance B-cell but not T-cell immune responses and cannot protect mice from tumor challenge.
