一测多评法测定柿叶提取物中6种黄酮类成分

摘    要:目的 建立一测多评(quantitative analysis of multi-components by single-marker,QAMS)法同时测定柿叶提取物中6种黄酮类成分的含量。方法 采用Agilent Eclipse Plus C18色谱柱(250 mm×4.6 mm,5μm),流动相为乙腈-0.1%磷酸水溶液,梯度洗脱,流速1.0 mL·min-1,检测波长360 nm,柱温30℃,进样量10 μL。应用QAMS法,以槲皮素为内参物,测定5种黄酮类成分之间的相对校正因子,分别采用外标法和QAMS法测定柿叶提取物中的6种指标性成分的含量,并对2种方法计算结果进行对比分析,以验证QAMS法的实用性与稳定性。结果 建立了用于测定柿叶提取物中6种黄酮类成分的QAMS法,各成分相对校正因子的稳定性和重复性良好,QAMS法和外标法的测定值差异较小。结论 本研究建立的一测多评法测定柿叶提取物中6种黄酮类成分,方便快捷,结果准确可靠,可用于柿叶提取物多指标成分的质量控制。

Abstract:OBJECTIVE To develop a method of quantitative analysis of multi-components by single-marker(QAMS) for simultaneous determination of six flavonoids in extract of Diospyros kaki Thunb. leaves. METHODS The Agilent Eclipse Plus C18 column(250 mm×4.6 mm, 5 μm) was applied, the mobile phase was acetonitrile-0.1% phosphoric acid of gradient elution, the flow rate was 1.0 mL·min−1, the detection wavelength was 360 nm, the column temperature was 30 ℃ and the injection volume was 10 mL. Used quercetin as a reference, the relative correction factors among the five flavonoids were detected by QAMS. The content of six index components in extract of Diospyros kaki Thunb. leaves. was determined by external standard method and QAMS. The results obtained from two different methods were compared, verified the practicability and stability of QAMS. RESULTS The QAMS was established and used to determine the six flavonoids in the extract of Diospyros kaki Thunb. leaves. The results showed that the relative correction factors had good system applicability and reproducibility, and there was no significant difference between method QAMS and external standard method. CONCLUSION The established QAMS method for simultaneous determination of six flavonoids in extract of Diospyros kaki Thunb. leaves is simple, accurate, reproducible and feasible to quantify multiple components in extract of Diospyros kaki Thunb. leaves.
