明党参根皮中5种呋喃香豆素类成分的体外抗肿瘤活性

目的:研究明党参根皮中异欧前胡素、欧前胡素、花椒毒酚、珊瑚菜内酯、5-羟基-8-甲氧基补骨脂素5种呋喃香豆素类成分的体外活性.方法:选取人肝癌细胞株SMMC-7721,HepG2,人肺癌细胞株A-549,人胃癌细胞株MKN-45,人官颈癌细胞株Hela,人乳腺癌细胞株MCF-7,MDA-MB-2317种肿瘤细胞.以5×104个/mL的细胞密度接种于96孔板,给予不同浓度的药物后培养72 h,采用MTT法观察5种成分对各细胞活性的影响.结果:5种呋喃香豆素对7种肿瘤细胞均表现出不同程度的增殖抑制作用,IC50在0.30～17.23 mg·L-1之间,其中异欧前胡素效果最显著,IC50从0.39 mg·L-1到4.11 mg·L-1.结论:明党参根皮中5种呋喃香豆素类成分具有明显的抗肿瘤活性.

Study on Antitumor Activity of Five Furanocoumarins from the Root Bark of Changium smyrnioides in vitro

Objective: To study the in vitro activity of five furanocoumarins from the root bark of Changium smyrnioides Wolff. isoimperatorin, imperatorin, xanthotoxol, phellopterin and 5-hydroxyl-8-methoxy-psoralen. Method: The antitumor effect against human liver cancer (SMMC-7721 and HepG2), human lung cancer (A549), human gastric cancer (MKN-45), human uterine cervix cancer (Hela) and human breast cancer (MCF-7 and MDA-MB-231) was observed by MTT. Result: Five furanocoumarins had different inhibition on seven kinds of tumors. IC₅₀ was from 0.30 mg·L^-1 to 17.23 mg·L^-1, especially isoimperatorin was most significantly effective. IC₅₀ was from 0.39 mg·L^-1 to 4.11 mg·L^-1. Conclusion: These five furanocoumarins have obvious antitumor effect.
